A recombinant protein can be tagged N-terminally or C-ternimally with a 6XHis tag. This tag is better in prokaryotic system rather than mammalian system. 6X His tag is an affinity tag which allows the tagged recombinant protein to be purified in the process of affinity purification. 6XHis tag binds to NTA-agarose. The resin contains nickle ions which binds to Histidine tag.
